Will this adapter work on my desktop's 3.5" HDD?

Unfortunately, this adapter will only work on 2.5" drives. 3.5" HDDs require a 12V power for their spindle motors, which USB does not provide.

Will this USB to SATA adapter allow me to read the S.M.A.R.T. data of the drive?

As long as the drive you use supports S.M.A.R.T. data monitoring, yes it will.

Will this work with Thunderbolt 3 and allow up to 10Gbps with the included USB-A to USB-C adapter?

Yes, it can, as long as the host system supports the capability.
